Tries from Eli Walker, Bowdy Davies and Luke Morgan earned the visitors a 28-16 victory at Newbridge when the sides met in early September.
Since then, the Dragons have picked up their first victory of the season - 36-0 at Gogledd Cymru - whilst the Ospreys saw their unbeaten record disappear when the Blues won 21-6 at Llandarcy.
With the Blues currently on top of the pile, after three successive victories, both sides are desperate for a victory if only to ensure a runners up spot and a place in the final later in the season.
Dragons team manager Billy Vyvian said: "The boys have shown great spirit all season and would love to reverse the result of the last game we played against them.
"It's a difficult place to go, but confidence is fairly hiogh after a decent win in North Wales where we picked up a bonus point too."
Walker and Davies will pose the biggest threat to the Dragons - the pair scored a try apiece against the Dragons at Newbridge with Walker running in one of his side's four tries in the 37-12 victory over Gogledd Cymru in Colwyn Bay.
